As mentioned in the recent SCCS XI<\/strong>, the 11th revision<\/a><\/strong> of the \u201cGuidance Notes for the Testing of Cosmetic Ingredients and the Evaluation of their Safety<\/em>\u201d, in paragraph 3<\/span>\u2013<\/span>4.6, when talking about New Approach Methodologies (NAMs) for serious eye damage and eye irritation:<\/span><\/p>\n

TG OECD 492 <\/strong>uses a RhCE tissue model based on a human tissue culture.<\/p>\n

TG OECD 491<\/strong> uses a rabbit corneal cell line.<\/p>\n

TG OECD 437<\/strong> and 438<\/strong> use eyes collected from chickens and bovines obtained from slaughterhouses.<\/p>\n

TG OECD 460 <\/strong>makes use of kidney cells.<\/p>\n

Only Ocular Irritection\u00ae OECD TG 496<\/strong> is<\/strong> completely, 100%,<\/strong> animal free.<\/strong><\/span>

In fact, Ocular Irritection\u00ae<\/a><\/strong> is based on the knowledge that the ability of a chemical to irritate the cornea is related to its propensity to promote the denaturation and destruction of corneal proteins<\/strong>. <\/p>\n

The Assay System mimics these biochemical phenomena by using a protein reagent in chemico<\/em>.<\/p>\n

When the assay is performed, the changes in protein structure induced by the test sample are then quantified by measuring the resulting changes in turbidity <\/strong>(optical density) of the solution reagent at a wavelength of 405 nm using a spectrometer.<\/p>\n

The results of past comparative studies were used to develop what is called an “Irritection Draize Equivalent<\/strong>” (IDE) score, which is a value derived from in vitro<\/em>studies that is equivalent to the expected irritation potential in vivo (Draize rabbit eye test).<\/p>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t

Ocular Irritection\u00ae, when used by itself, is the first and only step in a top-down or bottom-up strategy for, respectively, the classification of substances that cause severe ocular damage (UN GHS Category 1<\/strong>) and that do not require classification (UN GHS No Category<\/strong>).<\/p>\n

The SCCS XI, in addition to the guidance document OECD 263<\/a><\/strong>, mentions the use of Ocular Irritection\u00ae even if the test substance is not UN Cat.1 or No Cat.<\/strong>: the top-down or bottom-up approach of OI\u00ae becomes part of an integrated strategy (IATA –<\/strong> Integrated Approach on Testing and <\/i>Assesment<\/i>) and one of the many steps to take within it.<\/p>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t
